I started running at about 260 to 265 lbs. Keep your pace slow like others have said, and make sure you get fitted for proper running shoes. I'd have to recommend SL5x5. It's a compound lifting program (not isolation), so it'll work the majority of your muscles. This is of course if you don't have anything already planned.

I do Stronglifts 5x5 Mon, Wed, and Fri followed by C25K Tues, Thurs, and Sat. However, my back is acting up so I'm only doing bench and OHP in Stronglifts right now.

I have incredibly depressing and cranky coworkers on my team. I just walk around with a **** eating grin, and a belly laugh. It also helps that I'm really quite awesome, and remind them on a regular basis of that. They warmed up pretty fast.

SL5x5 is great for building strength. I did about 16 weeks of it, and then stopped to focus more on C25K for a while, and now I'm getting back into it (with a 50% de-load). It's meant for building strength, and focuses on compound lifting instead of isolation.
