A letter is always a bonus. I might have 50+ candidates for a position, interview 5-10, and by the time I get to the interviews, I really am pressed for time. A letter for me would impress me much more than a follow up phone call.
I would like to hear a thank you for my time in interviewing you, what you would look forward to about working for the company, something to show you've looked into the company a little (a little online research about the place never hurts), and why your skills would fit well with the position. I would expect that you would reiterate that you are very interested in the position and hope to hear from me soon about the position.
In my field it might look something like:
"Thank you for taking the time to speak with me Tuesday about the open (whatever) position with your company, and after discussing the skills required, I am even more certain the experience I've gained in (your experience here) would be an asset to your company.
After learning about the positive impact (Business) has on patients' lives, I am even more excited about working with your team in the (wherever) office. It is exciting to hear about the recent growth of the company, particularly with the expansion into the field of (you get the idea).
Thank you again for the opportunity to interview for the position, and I look forward to hearing from you very soon.
Sincerely,"
Obviously, tweaked to fit. But something along those lines, I would LOVE to see from an applicant after an interview and it would definitely bump you up on my 1-10 scale.0 -

Ok good, the application was very minimal. It basically asked me to upload a resume and them in pulled some info out of my resume. Is it ok to ask if they would like references or if I should provide a letter of recommendation or anything else?0

Just a thought too.... depending on the size of the company, if it's a large company and you applied online, your application may first go to a separate department or a recruiter before it's sent to the place where the position is for which you are actually applying. If it's a large company, your follow up application call might be best addressed with "Hi, my name is ____, and I just applied online. I just wanted to check if the position is still open and to let you know I'm very interested in the position."
