How do I know if I've lost muscle mass of fat?

Unless you get dexa scans or bodpod every 6 months or so you won't really know. Other BF testing can give you a rough estimate, but can be very unreliable.
Almost all weight loss will result in some amount of LBM loss. The usual recommendations to preserving as much LBM as possible are getting adequate protien (I forget the % you should aim for), lifting, and a reasonable deficit for what your looking to lose.0 -
80 -100 grams of protein a day is usually recommended, as is incorporating some sort of strength training. It doesn't have to be lifting weights - doing exercises that incorporate body weight like squats, lunges, push-ups & planks can help you retain muscle mass while losing weight.0
