When working out on the treadmill it registers different calories burned than MyFitness. Today I walked on an incline level of 8 @ 2.5mph for 45min. When I was done Treadmill registered 450 calories burned MyFitness registered 144. Do I manually add the difference?

I wouldn't. MyFitnessPal is probably closer to correct on that one.2

I agree. MFP is far from perfect, but I don't trust any machine's calorie count. I would start logging with the MFP calculations, and if you are not losing weight at your desired rate, you can reassess. If you are losing too slowly and you feel confident you are logging your food correctly, some people will only eat 75-50% of MFP's calories back.
I started out using MFP's calorie burns for my walking and ate back 100% of them and lost weight. So just try out logging the MFP calories and seeing if you are getting the results you want after a few weeks.0
