Do you have to do squats with weights to see a difference? ??

if you are beginner then no, you can just start doing body weight squats.
However, at some point you are going to want to progress to weighted squats like goblet squats, or barbell squats. Additionally, I would highly recommending getting into barbell squats as it is an awesome workout that works your entire body….
