Reading the links in the thread below (especially the "Step by step" one) is a great place to start. After that, go through MFP's guided setup, enter your stats, and choose a reasonable weekly weight loss goal. Generally speaking, if you have:
100+ lbs to lose: choose 2 lb/week
75+ lbs to lose: choose 1.5 lb/week
Less than 60-70 lbs to lose: choose 1 lb/week.
MFP will then give you a daily calorie goal. After that, buy a food scale if you don't already have one. Commit to weighing every single thing you eat (including packaged foods). Use measuring cups only for actual liquids like juice, milk, etc. Log everything into your daily food diary. Use the recipe builder (under Food/Recipes) if you cook a lot a home. After 4-6 weeks, reassess your progress and make some adjustments if necessary. And of course, always feel free to post in the forums if you have any other questions.
Good luck!
