Does it take for calories you ate to show up on your body?

Let's say, hypothetically, that on maintenance I need 2000 calories per day. One day, I decide to eat nothing but deep fried mac and cheese, and cheesecake and end up eating 5500 calories. Will my body turn all of those calories into fat that day? The next day? The next week? Is this something that we can even measure?
